Abstract

Objective: Evaluation of prognostic factors associated with survival outcomes in patients with metastatic pancreatic cancer receiving first-line mFOLFIRINOX.

Methods: A retrospective and prospective study on 36 patients diagnosed with metastatic pancreatic cancer at K Hospital were treated with the first-line mFOLFIRINOX regimen. The primary endpoints were overall survival (OS) and progression-free survival (PFS), with analyses of factors associated with OS and PFS. Secondary endpoints included response rate, treatment-related toxicity, and the rate of treatment interruption.

Results: The median follow-up was 15.6 months (95% CI, 3.8–27.3). The median progression-free survival was 6.7 months, and the median overall survival was 12.5 months. In univariate analysis, age <60 and ECOG performance status (PS) 0 were associated with prolonged progression-free survival (PFS) and overall survival (OS). ECOG PS 0 was identified as an independent prognostic factor for overall survival. The overall response rate (ORR) was 30.6 %, disease control rate (DCR) was 66.6%. Grade 3 or 4 toxicities were mainly hematologic, including neutropenia in 33.4% of patients and thrombocytopenia in 2.8%

Conclusion: The mFOLFIRINOX regimen demonstrated treatment efficacy. ECOG PS 0 was identified as favorable prognostic factors associated with prolonged survival in patients with metastatic pancreatic cancer.
